Some friends and myself are looking to play some DND 3.5 or 4.0 online, and I believe I remember seeing a few programs posted that would give us the freedom to do this.
I can't remember any of the names, though :/

Also, Wizards of the Coast has a series of tools called D&D Insider (http://www.wizards.com/dnd/insider/") that, when fully released (some tools are in beta right now) will allow you to build characters, check rules, and play online.
EDIT: Most of the D&D Insider tools do require a subscription, though.

I know it's not quite the same thing, but I would also like to recommend Apprentice (http://www.magic-league.com/download/apprentice.php), a free program that lets you play Magic: The Gathering online on a virtual table. My friends and I use it a lot when I'm away.
